Compact Cell Imaging Device (CoCID) provides insights into the cellular origins of viral infections

The overall CoCID concept is centred on providing virologists with a next-generation imaging device, which, through increased penetration and depth of focus, as well as through high natural contrast and sensitivity to organelle density (including virus-related organelles), will produce higher-fidelity ultrastructural images of whole intact cells. These insights will, in turn, help increase our understanding of the links between the structural reorganisation of cells and the mechanisms of viral entry, replication, assembly, and egress in cells. Soft X-ray emission spectroscopy used for the characterization of a-C and CNx thin films

Abstract We present the results of a soft X-ray emission spectroscopy study of a-C and CNx films on a Si(100) substrate. Also for the characterization of the homogeneity in depth of these films electron energy loss spectroscopy measurements with localization better than 4 nm were carried out. In case of CNx films the highest diamond-like modification occurs in the region close to the Si(100) substrate. The film density decreases with increasing distance from the substrate and becomes almost constant in range of thicknesses more than ~ 2 nm.

Each class of microscope is limited to imaging specific aspects of cell structure and/or molecular organization. However, imaging the specimen by complementary microscopes and correlating the data can overcome this limitation. Whilst not a new approach, the field of correlative imaging is currently benefitting from the emergence of new microscope techniques. Here we describe the correlation of cryogenic fluorescence tomography (CFT) with soft X‐ray tomography (SXT). This amalgamation of techniques integrates 3D molecular localization data (CFT) with a high‐resolution, 3D cell reconstruction of the cell (SXT). Study and implementation of a soft X-ray 100 eV -20 keV fixed exit monochromator system

We describe a “built in house” X-ray monochromator which produces a fixed exit X-ray beam tunable in the full energy range 0.1 - 20 keV. The system is based on a double diffraction on two large size parallel crystals positioned using a remotely controlled micropositioning system in order to keep the position of the monochromatic beam for any chosen energy. Up to six different diffracting elements can be selected without breaking the vacuum. This allows to cover the full energy range of interest. Second order reflection from crystals used in soft X-ray spectroscopy

Abstract In this note the ratio of the second to the first order reflection is determined for the KAP and PbSt crystals, for wavelengths corresponding to the Al K-line emission. The source of the radiation was a low-voltage stabilized X-ray tube. The X-rays were detected with a Bragg spectrometer equipped with a proportional counter detector. The signal measured by the proportional counter was subsequently pulse height analyzed.

Upon infection, viruses hijack the cell machinery and remodel host cell structures to utilize them for viral proliferation. Since viruses are about a thousand times smaller than their host cells, imaging virus-host interactions at high spatial resolution is like looking for a needle in a haystack. Scouting gross cellular changes with fluorescent microscopy is only possible for well-established viruses, where fluorescent tagging is developed.
